fa ارزیابی اثر محیط‌کشت و ترکیبات هورمونی مختلف بر پارامترهای درون شیشه‌ای در طی ریزازدیادی ارکیده وانیلی (Vanilla planifolia) Evaluation of the effect of culture medium and different hormonal compounds on in vitro parameters during the micropropagation of vanilla orchid (Vanilla planifolia) باغبانی Horticulture پژوهشي Research در این مطالعه یک روش کارآمد و ساده برای ریزازدیادی ارکیده وانیلی (Vanilla planifolia) که یک گیاهی ادویهای- زینتی ارزشمند است توسعه داده شد. ریزنمونههای میانگره شاخه پس از فرآیند ضدعفونی روی هشت محیطکشت شامل MS + 2 میلیگرم در لیتر TDZ؛ MS + 2 میلیگرم در لیتر TDZ + 5/0 میلیگرم در لیتر NAA؛ MS + 2 میلیگرم در لیتر Kin؛ MS + 2 میلیگرم در لیتر Kin + 5/0 میلیگرم در لیتر NAA؛ MS2/1 + 2 میلیگرم در لیتر TDZ؛ MS2/1 + 2 میلیگرم در لیتر TDZ + 5/0 میلیگرم در لیتر NAA؛ MS2/1 + 2 میلیگرم در لیتر Kin؛ MS2/1 + 2 میلیگرم در لیتر Kin + 5/0 میلیگرم در لیتر مستقر شدند. تیمارها در قالب طرح کاملاً تصادفی در سه تکرار آرایش یافتند و هر ماه یکبار واکشت انجام شد. پس از سه ماه و اتمام مرحله ریزافزایی صفات مختلف مورد ارزیابی قرار گرفت. نتایج نشان داد که روی محیط 1/2MS+2 mg/l TDZ+0.5 mg/l NAA و 1/2MS+2 mg/l Kin+0.5 mg/l NAA بیشترین میزان زندهمانی ریزنمونه (91 درصد) و شاخهزایی رخ میدهد. با این حال، محیط1/2MS+2 mg/l TDZ+0.5 mg/l NAA  منجر به بیشترین تعداد برگ، وزن تر و درصد وزن خشک نیز شد و همچنین بالاترین تعداد ریشه و وزن تر ریشه نیز روی این محیط مشاهده شد. طول گیاهچههای ریزافزایی شده مستقر روی محیطکشت MS+2 mg/l TDZ+0.5 mg/l NAA بیشتر از سایر محیطهای کشت بود. بهطورکلی، براساس نتایج بدست آمده محیط 1/2MS+2 mg/l TDZ+0.5 mg/l NAA برای ریزازدیادی ارکیده وانیلی پیشنهاد میشود.  In this study, an efficient and simple method was developed for the micropropagation of vanilla orchid (Vanilla planifolia), which is a valuable spice-ornamental plant. The internodal explants after the disinfection process were established on eight culture mediums, including MS + 2 mg/L TDZ; MS + 2 mg/L TDZ + 0.5 mg/L NAA; MS + 2 mg/L Kin; MS + 2 mg/L Kin + 0.5 mg/L NAA; 1.2MS + 2 mg/L TDZ; 1.2MS + 2 mg/L TDZ + 0.5 mg/L NAA; 1.2MS + 2 mg per liter Kin; 1.2 MS + 2 mg/l Kin + 0.5 mg/L NAA. The treatments were arranged in the form of a completely randomized design (CRD) in three replications, followed by subculturing once every month. After three months and the completion of the shoot proliferation stage, different in vitro traits were evaluated. The results showed that the highest survival rates of explants and shoot multiplication occur on <a name="_Hlk144401594">1/2MS+2 mg/l TDZ+0.5 mg/l NAA </a>and 1/2MS+2 mg/l Kin+0.5 mg/l NAA mediums. However, 1/2MS+2 mg/l TDZ+0.5 mg/l NAA medium resulted in the highest leaf number, fresh weight and percentage of dry weight. The length of tissue-cultured plants on MS+2 mg/l TDZ+0.5 mg/l NAA was greater than other mediums. The highest number of roots and root fresh weight were also observed at 1/2MS+2 mg/l TDZ+0.5 mg/l NAA, although compared to other mediums with NAA there was not a significant difference.
