Given the medicinal value, there is a need to develop effective strategies to enhance the essential oil yield and strengthen the physiological responses of Dracocephalum kotschyi. On the other hand, the mechanisms of action of elicitors such as titanium dioxide nanoparticles (TiO2) and methyl jasmonate (MeJA) on this plant remain unknown. Therefore, this study aimed to investigate the individual and combined effects of these elicitors on the physiological response, the expression of limonene biosynthesis genes, and the essential oil yield in D. kotschyi. After seedlings were established in plastic pots, TiO2 (at concentrations of 100 and 200 mg/L) and MeJA (at a concentration of 0.5 mM) were sprayed separately and in combination onto the D. kotschyi plants in three rounds at three-week intervals. Sampling was performed 48 h after the last spraying. The results showed that both elicitors, individually and in combination, affected the traits in a concentration-dependent manner. The combined application of TiO2 nanoparticles acted synergistically to improve the traits of D. kotschyi. In particular, the treatment with 0.5 mM MeJA along with 200 mg/L TiO2 led to an increase in the activity of antioxidant enzymes (superoxide dismutase, catalase, and polyphenol oxidase), soluble sugars, and carotenoids. Furthermore, this treatment enhanced the expression of the key genes GPPS and LS involved in limonene biosynthesis by 3.9- and 3.4-fold, respectively, and increased the essential oil yield by 3.6-fold compared to the double control. Overall, this combined treatment, by creating a synergistic effect, improved the activity of the antioxidant system, osmolytes, the expression of key genes in limonene biosynthesis, and the essential oil yield of D. kotschyi.
